BDP FL NHS ester is an advanced dye for 488 nm channel, a replacement for fluorescein, a molecule identical to BODIPY FL? NHS ester. An amino-reactive dye for the labeling of proteins and peptides.

While the absorbance and emission spectra of this molecule stay within FAM excitation and emission channels, this dye provides much better photostability, and outstanding brightness. The fluorescence spectrum of BODIPY-FL is narrower than that of FAM. This provides a better brightness for monochromator based instruments, when emission wavelength can be tuned to dye maximum.

The dye is neutral, possesses low molecular weight, and retains high quantum yield in conjugates.

The dye is a good replacement for fluorescein (FAM), BODIPY-FL, Alexa Fluor 488, DyLight 488, Cy2, and other 488 nm dyes.

Bodipy類熒光染料具有良好的穩(wěn)定性、優(yōu)良的光學(xué)性能,主要表現(xiàn)在以下幾個(gè)方面:

1:BODIPY的光譜峰寬比較窄,在光學(xué)領(lǐng)域和生物學(xué)領(lǐng)域進(jìn)行分析檢測(cè)時(shí),光譜峰寬較窄,檢測(cè)靈敏度較高,并且減少了待測(cè)樣品的用量。測(cè)得的結(jié)果中,熒光光譜上會(huì)出現(xiàn)更為明顯的熒光吸收和發(fā)射的變化,這樣更精確地測(cè)得了樣品的含量。

2:在整個(gè)BODIPY分子中,并不存在負(fù)電荷。因而在溶液中,溶液中的離子無(wú)法與缺少電荷的BODIPY分子發(fā)生靜電作用,這樣非常有效的避免了測(cè)定時(shí)靜電作用對(duì)測(cè)量結(jié)果的干擾,使測(cè)定過(guò)程更加穩(wěn)定,測(cè)得結(jié)果更加精準(zhǔn)。

3:BODIPY類熒光染料的光穩(wěn)定性能非常優(yōu)越,當(dāng)它被激發(fā)到達(dá)激發(fā)態(tài)時(shí),不會(huì)因?yàn)楹荛L(zhǎng)時(shí)間的激發(fā)而在分析測(cè)定過(guò)程中發(fā)生激發(fā)態(tài)的分解或其他變化,這樣測(cè)得的熒光光譜更加準(zhǔn)確可信。

4:BODIPY染料,其熒光量子產(chǎn)率甚至接近1.0。一般普通的染料在水溶液中其熒光量子產(chǎn)率會(huì)變低,有的甚至被猝滅。對(duì)于BODIPY分子來(lái)說(shuō),在水溶液中其熒光量子產(chǎn)率雖然有所下降,但它仍然能保持較高的熒光量子產(chǎn)率。

5:由BODIPY的母體結(jié)構(gòu)可知,他的母體結(jié)構(gòu)上的取代基一般不會(huì)出現(xiàn)氨基或羧基這類對(duì)酸堿敏感的活性基團(tuán)。因此外界的酸堿度對(duì)其影響會(huì)大幅度減小,因此BODIPY染料不會(huì)輕易變質(zhì)。

6:BODIPY類熒光染料對(duì)光的感應(yīng)度很高,并且它還有高的摩爾消光系數(shù),有些經(jīng)過(guò)修飾后的BODIPY染料在水溶液中的溶解度也較高,再加上它在水中不易被猝滅這一優(yōu)異的性能,使得BODIPY在光學(xué)、生物學(xué)、分析學(xué)領(lǐng)域有著極其廣泛的應(yīng)用。

BODIPY母體結(jié)構(gòu)最初是由德國(guó)化學(xué)家Treibs, A.和kreuzer, F. H.在1968年合成的,他們合成的BODIPY 類熒光染料 [40] 。另外,他們還合成了BODIPY染料的母體結(jié)構(gòu)。在BODIPY母體結(jié)構(gòu)中,Treibs, A.和kreuzer, F. H.在R1-R7中用不同基團(tuán)取代得到不同的衍生產(chǎn)物。人們根據(jù)Treibs, A.和kreuzer, F. H.的前期探索,對(duì)BODIPY類染料做出了多種結(jié)構(gòu)修飾,這大大豐富了BODIPY的種類,使其迅猛發(fā)展。
