DiD(CAS:127274-91-3),DiO(CAS:34215-57-1),DiI(CAS:41085-99-8),DiR(CAS:100068-60-8)和DiS染料是一族親脂性的熒光染料,可以用來染細(xì)胞膜和其它脂溶性生物結(jié)構(gòu)。當(dāng)與細(xì)胞膜結(jié)合后其熒光強(qiáng)度大大增強(qiáng),這類染料有著很高的淬滅常數(shù)和激發(fā)態(tài)壽命。一旦對(duì)細(xì)胞染色,這類染料在整個(gè)細(xì)胞膜上擴(kuò)散,佳濃度時(shí)可以使整個(gè)細(xì)胞膜染色。它們的熒光顏色區(qū)分明顯:DiI(橙色熒光),DiO(綠色熒光),DiD(紅色熒光)和 DiR (深紅色熒光)這使得他們可以用來對(duì)活細(xì)胞進(jìn)行多色成像和流式分析。DiI和DiO可以分別用標(biāo)準(zhǔn)的 FITC和TRITC的濾光片。DiD可以用 633 nm He–Ne 激光器激發(fā),有著比DiI更長(zhǎng)的激發(fā)波長(zhǎng)和發(fā)射波長(zhǎng),在細(xì)胞和組織染色中更有價(jià)值。 DiR的紅外熒光可以穿透細(xì)胞和組織,在活體成像中用來示蹤。

**DiR碘化物的發(fā)射波長(zhǎng)肉眼不可見,所以需要配備CCD鏡頭或其他的近紅外檢測(cè)儀器。

Di系列染料想必大家一定不陌生,我們做細(xì)胞膜染色的時(shí)候經(jīng)常會(huì)用到,今天給大家介紹下常見的4類Di染料:DiI, DiO, DiD 和 DiR,Di系列染料是一類環(huán)境敏感型親脂性膜染料,這類探針在進(jìn)入細(xì)胞膜之前熒光非常弱,當(dāng)它們與膜結(jié)合或者與親脂性生物分子(例如蛋白質(zhì))結(jié)合時(shí),其熒光強(qiáng)度顯著增強(qiáng)。一旦應(yīng)用于細(xì)胞中,這類染料會(huì)在細(xì)胞內(nèi)質(zhì)膜中逐步擴(kuò)散,在其最佳濃度條件下,可以擴(kuò)散逐漸使整個(gè)細(xì)胞的細(xì)胞膜被染色。

 

DiI, DiO, DiD 和 DiR被激發(fā)后可以發(fā)出不同顏色的熒光,DiO和磷酯雙層膜結(jié)合后的激發(fā)光譜和發(fā)射光譜參考下圖。

其中I、D、R分別代表了一個(gè),兩個(gè),三個(gè)雙鍵,它們是決定波長(zhǎng)長(zhǎng)度的關(guān)鍵,DiO, DiI, DiD 和 DiR這幾款染料的結(jié)構(gòu)式如下:

關(guān)于染料的使用技巧:

1.DiI 可以溶解于無水乙醇、DMSO 和 DMF,溶解度約為 1-2.5mg/ml,發(fā)現(xiàn)較難溶解時(shí)可以適當(dāng)用超聲處理以促進(jìn)溶解。

2.用于細(xì)胞膜熒光標(biāo)記時(shí),DiI 的常用濃度為 1-25μM,最常用的濃度為 5-10 μM。

3.?DiI 可以直接染色活的細(xì)胞或組織,染色時(shí)間通常為 5-20 分鐘。

4.?染固定的細(xì)胞時(shí),通常使用 4%多聚甲醛進(jìn)行固定,使用其它不適當(dāng)?shù)墓潭ㄒ簳?huì)導(dǎo)致熒光背景較高。

DiD,DiO,DiI,DiR熒光染料之間的區(qū)別

DiD染料是一族親脂性的熒光染料,可以用來染細(xì)胞膜和其它脂溶性生物結(jié)構(gòu)。當(dāng)與細(xì)胞膜結(jié)合后其熒光強(qiáng)度大大增強(qiáng),這類染料有很高的淬滅常數(shù)和激發(fā)態(tài)壽命。一旦對(duì)細(xì)胞染色,染料在整個(gè)細(xì)胞膜上擴(kuò)散,zui佳濃度時(shí)可以使整個(gè)細(xì)胞膜染色。

綠色熒光、親脂性羰花青DiOC18(3)被廣泛用作親脂示蹤劑。它在水里是弱熒光的,但當(dāng)嵌入膜時(shí),它的熒光和光穩(wěn)定性都很好。

DiI是一種親脂性膜染料,進(jìn)入細(xì)胞膜后可以側(cè)向擴(kuò)散逐漸使整個(gè)細(xì)胞的細(xì)胞膜被染色。DiI在進(jìn)入細(xì)胞膜之前熒光非常弱,僅當(dāng)進(jìn)入到細(xì)胞膜后才可以被激發(fā)出很強(qiáng)的熒光。DiI被廣泛用于正向或逆向的,活的或固定的神經(jīng)等細(xì)胞或組織的示蹤劑或長(zhǎng)期示蹤劑,通常不會(huì)影響細(xì)胞的生存力。

DIR是一個(gè)親脂性、近紅外熒光花青染料。這個(gè)染料常用于標(biāo)記細(xì)胞質(zhì)膜。兩個(gè)18-碳鏈插入到細(xì)胞膜,從而特定的、穩(wěn)定的細(xì)胞染色,幾乎不會(huì)發(fā)生細(xì)胞間的染料轉(zhuǎn)移??梢杂靡掖寂渲闷鋬?chǔ)存液。細(xì)胞染色通常使用的濃度時(shí)1-10uM,孵育10-20分鐘的。

使用方法
1.?DiD,DiO,DiI,DiR和DiS細(xì)胞膜染色液制備
(1)配置DMSO或EtOH?儲(chǔ)存液:儲(chǔ)存液用?DMSO或EtOH配置濃度1~5?mM。
注意:未使用的儲(chǔ)存液保存在-20?oC,避免反復(fù)凍融。
(2)工作液制備:用合適的緩沖液(如:無血清培養(yǎng)基,HBSS或PBS)稀釋儲(chǔ)存液,配制濃度為1~5?μM的工作液。
注意:工作液的最終濃度是根據(jù)不同細(xì)胞和實(shí)驗(yàn)的經(jīng)驗(yàn)來配制??梢詮耐扑]濃度的十倍以上尋找佳條件。
2.?懸浮細(xì)胞染色
(1)懸浮細(xì)胞密度為?1?×?106/mL加入到工作液中。
(2)在37 ℃培養(yǎng)細(xì)胞 2~20分鐘,不同的細(xì)胞佳培養(yǎng)時(shí)間不同。
(3)染色細(xì)胞試管在1000~1500轉(zhuǎn)離心5分鐘。
(4)傾倒上清液,再次緩慢加入預(yù)溫37℃的培養(yǎng)液。
(5)重復(fù)(3),(4)步驟兩次以上。
3.?粘壁細(xì)胞的染色
(1)使粘壁的細(xì)胞在無菌實(shí)驗(yàn)室培養(yǎng)。
(2)從培養(yǎng)基中移走蓋玻片,吸走過量培養(yǎng)液,將蓋玻片放在潮濕的環(huán)境中。
(3)在蓋玻片的一角加入100μL的染料工作液,輕輕晃動(dòng)使染料均勻覆蓋所有細(xì)胞。
(4)在37 ℃培養(yǎng)細(xì)胞 2~20分鐘,不同的細(xì)胞佳培養(yǎng)時(shí)間不同。
(5)吸干染料工作液,用培養(yǎng)液洗蓋玻片2~3次,每次用預(yù)溫的培養(yǎng)基覆蓋所有細(xì)胞,培養(yǎng)5~10分鐘,然后吸干培養(yǎng)基。
4.?顯微鏡檢測(cè)
(1)DiD,DiO,DiI,DiR和DiS濾光器的選擇參見表1。
(2)多色染料的同時(shí)檢測(cè),濾光器按照以下設(shè)定:
a)?DiI和DiO=Omega?XF52,Chroma?51004;
b)?DiI和DiD=Omega?XF92,Chroma?51007;
c)?DiI,DiO和DiD=Omega?XF93,Chroma?61005
5.?流式細(xì)胞儀的檢測(cè)
DiD,DiO,DiI,DiR和DiS?染色的細(xì)胞可以分別用經(jīng)典的FL1,F(xiàn)L2,F(xiàn)L3和FL4流式細(xì)胞儀檢測(cè)。

The kit is used for the preparation of labeled antibodies having 2.5-3 labels per antibody. The kit contains all reagents and materials for ten labeling reactions, each for 100 ug of the antibody. The dye is an NHS ester, taken in controlled excess. It reacts with the amine groups of lysines the antibody at mild alkaline pH. The purification of the antibody is achieved by gel filtration using spin columns. These kits contain sulfonated, water soluble dyes, that are most suitable for the labeling of sensitive proteins, including antibodies.

Protocol

1. Antibody preparation.

Optimal conditions for the labeling reaction are the following: antibody at a concentration of 1 mg/mL dissolved in 0.1 M sodium hydrocarbonate solution. Sodium azide is compatible with the labeling. If concentration of antibody is below 1 mg/mL, it should be concentrated. Antibody preparation should be free of aminoacids, proteins like BSA and buffer components that bring pH to unfavourable (2-7.5 or 9-12) ranges.

2. Setting up the reaction.

2.1. Add a solution of antibody in sodium hydrocarbonate (100 ug in 100 uL*) to the fluorophore, vortex, and incubate for 30 min at room temperature.

* if a smaller amount of antibody is to be labeled, dissolve lyophilized dye in 10 uL of anhydrous DMSO, and take 1 uL of the solution per 10 ug of antibody. The solution of dye in DMSO should be consumed immediately.

3. Purification of the labeled antibody.

3.1. Prepare the column. The column should be at room temperature prior to use. The gel should be resuspended using vortex. Put the column into a waste receptacle (without cap), and centrifuge for 2 min at 1,000 g (speed must be carefully controlled – for standard 6 cm rotor, 1,000 g = 3800 rpm). The small bulge of the column should be oriented outwards.

3.2. Apply 400 uL of 1x PBS buffer, centrifuge for 2 min at 1,000 g.

3.3. Remove the column from waste receptacle. Put it into collection tube (with cap). Apply 100 uL of the reaction mixture onto the column, incubate for 1 min, and elute the antibody by centrifugation for 2 min at 1,000 g. Add 1/100 volume of 100x sodium azide. Antibodies can be aliquoted to improve their shelf life. Current aliquot can be stored at +4°С the rest at -20°С.

4. Measurement of dye to antibody ratio.

To calculate dye to antibody ratio, measure absorption spectrum of the conjugate, or absorption at 280 nm (AAB), and at dye absorption maximum (ADye). A typical absorption spectrum of a dye labeled antibody is shown below. Depending on the dye, wavelength of the maximum may vary.

An absorption spectrum of a labeled antibody contains dye peak (with longer wavelength), and antibody peak (at around 280 nm). Dye to antibody ratio is calculated using the following formula:

Example of calculation

After labeling of IgG antibody with sulfo-Cyanine5 and purification, a solution has been obtained with absorption spectrum above. Determine dye to protein ratio.

Using absorption spectrum, the following values can be found:?ADye?= 0.184 at dye absorption maximum (646 nm, see the table),?AAB?= 0.066 (at 280 nm),?εAB?= 210000 for IgG;?εDye?= 271,000, and?CF280?= 0.04.

Dye/AB?– is 2.43 fluorophore molecules per antibody.

Interpretation of results, and antibody storage

In most cases, optimal dye to antibody ratio is 2-3 dye molecules per antibody. Increase of dye loading above this value does not improve fluorescent signal because of the concentrational quenching of the fluorescence. When too few fluorophores are attached during the reaction, decrease antibody loading per reaction. Use of expired kit can also lead to this result.

We recommend to test binding of labeled antibodies with their antigens. The labeled antibodies can be stored at -20°С. An aliquot that is currently in use, should be stored at +4°С to avoid freeze-thaw cycles. The stability of the conjugate is determined by antibody itself rather than fluorophore or the chemical bond. Labeled antibodies should not be exposed to direct sunlight for extended time, but tolerate ambient lighting well.

BDP TR is a bright and photostable borondipyrromethene dye, which is especially suitable for microscopy applications. The dye has a long excited state lifetime, and is therefore is useful for fluorescence polarization assays. Its large two-photon cross section makes this dye useful for two-photon spectroscopy.

BDP 581/591 is a borondipyrromethene dye that has relatively long fluorescence lifetime and two photon excitation cross section. It is useful for fluorescence polarization assays. Due to the presence of a diene system, it can react with reactive oxygen species (ROS) with a change of fluorescence.

This is the NHS ester derivative for the conjugation with primary and secondary amine groups of proteins, peptides, and other molecules.

BDP FL NHS ester is an advanced dye for 488 nm channel, a replacement for fluorescein, a molecule identical to BODIPY FL? NHS ester. An amino-reactive dye for the labeling of proteins and peptides.

While the absorbance and emission spectra of this molecule stay within FAM excitation and emission channels, this dye provides much better photostability, and outstanding brightness. The fluorescence spectrum of BODIPY-FL is narrower than that of FAM. This provides a better brightness for monochromator based instruments, when emission wavelength can be tuned to dye maximum.

The dye is neutral, possesses low molecular weight, and retains high quantum yield in conjugates.

The dye is a good replacement for fluorescein (FAM), BODIPY-FL, Alexa Fluor 488, DyLight 488, Cy2, and other 488 nm dyes.

Bodipy類熒光染料具有良好的穩(wěn)定性、優(yōu)良的光學(xué)性能,主要表現(xiàn)在以下幾個(gè)方面:

1:BODIPY的光譜峰寬比較窄,在光學(xué)領(lǐng)域和生物學(xué)領(lǐng)域進(jìn)行分析檢測(cè)時(shí),光譜峰寬較窄,檢測(cè)靈敏度較高,并且減少了待測(cè)樣品的用量。測(cè)得的結(jié)果中,熒光光譜上會(huì)出現(xiàn)更為明顯的熒光吸收和發(fā)射的變化,這樣更精確地測(cè)得了樣品的含量。

2:在整個(gè)BODIPY分子中,并不存在負(fù)電荷。因而在溶液中,溶液中的離子無法與缺少電荷的BODIPY分子發(fā)生靜電作用,這樣非常有效的避免了測(cè)定時(shí)靜電作用對(duì)測(cè)量結(jié)果的干擾,使測(cè)定過程更加穩(wěn)定,測(cè)得結(jié)果更加精準(zhǔn)。

3:BODIPY類熒光染料的光穩(wěn)定性能非常優(yōu)越,當(dāng)它被激發(fā)到達(dá)激發(fā)態(tài)時(shí),不會(huì)因?yàn)楹荛L(zhǎng)時(shí)間的激發(fā)而在分析測(cè)定過程中發(fā)生激發(fā)態(tài)的分解或其他變化,這樣測(cè)得的熒光光譜更加準(zhǔn)確可信。

4:BODIPY染料,其熒光量子產(chǎn)率甚至接近1.0。一般普通的染料在水溶液中其熒光量子產(chǎn)率會(huì)變低,有的甚至被猝滅。對(duì)于BODIPY分子來說,在水溶液中其熒光量子產(chǎn)率雖然有所下降,但它仍然能保持較高的熒光量子產(chǎn)率。

5:由BODIPY的母體結(jié)構(gòu)可知,他的母體結(jié)構(gòu)上的取代基一般不會(huì)出現(xiàn)氨基或羧基這類對(duì)酸堿敏感的活性基團(tuán)。因此外界的酸堿度對(duì)其影響會(huì)大幅度減小,因此BODIPY染料不會(huì)輕易變質(zhì)。

6:BODIPY類熒光染料對(duì)光的感應(yīng)度很高,并且它還有高的摩爾消光系數(shù),有些經(jīng)過修飾后的BODIPY染料在水溶液中的溶解度也較高,再加上它在水中不易被猝滅這一優(yōu)異的性能,使得BODIPY在光學(xué)、生物學(xué)、分析學(xué)領(lǐng)域有著極其廣泛的應(yīng)用。
