bioGenousTM Red Blood Cell Lysis Solution是一款利用細胞內外存在鹽離子濃度差而導致細胞膜脹破的原理來裂解無核紅細胞的溫和型紅細胞裂解液。本產品經無菌處理，主要用于經酶消化分散的組織細胞的分離純化，淋巴細胞的分離純化以及組織細胞蛋白與核酸提取等實驗中紅細胞的去除。

使用說明

新鮮全血樣本：

1.         1倍體積的新鮮全血，加入3倍體積的紅細胞裂解液。如1mL新鮮全血加入3mL紅細胞裂解液，輕輕渦旋或顛倒混勻。

2.         冰上放置15分鐘，其間輕輕渦旋混勻兩次，紅細胞裂解后，溶液應該是清亮透明的。

3.         收集細胞：4℃，450g離心10 分鐘以沉淀白細胞，小心吸棄上清液。

4.         向白細胞沉淀中加入2倍起始血液體積的紅細胞裂解液，輕輕渦旋充分重懸白細胞。如起始血液為1mL，則加入2mL的紅細胞裂解液。

5.         4℃，450 g 離心10分鐘沉淀白細胞，小心并徹底吸去上清液。

6.         重懸細胞，用于后續(xù)實驗；如提取RNA，最好于此步開始使用DEPC水配制的溶液進行操作。

新鮮組織樣本：

1.         新鮮組織經膠原酶和蛋白酶（或腫瘤組織消化液K601003）消化分散成組織細胞懸液，離心棄去上清液，向沉淀中加入1mL紅細胞裂解液，輕輕渦旋或顛倒混勻。

2.         收集細胞：4℃，350g離心5分鐘以沉淀組織細胞，小心吸棄上清液。

3.         用PBS或基礎培養(yǎng)基重懸細胞，用于后續(xù)實驗。

注意事項：

本裂解液為無菌產品，分離細胞用于細胞培養(yǎng)時請注意無菌操作。為了您的安全與健康，請穿實驗服并戴一次性手套操作。
