本產(chǎn)品檢測(cè)靈敏度高,特異性強(qiáng),重復(fù)性好。多次重復(fù)檢測(cè)結(jié)果表明,最小檢出量為11.6pg/ml,與小鼠的FGF-basic,S100A13沒(méi)有交叉反應(yīng),與人FGF-acidic有21.9%的交叉反應(yīng)。板內(nèi)、板間變異系數(shù)均小于10%。
酸性成纖維細(xì)胞生長(zhǎng)因子(Fibroblast Growth Factor acidic, FGF-acidic),又稱FGF-1、內(nèi)皮細(xì)胞生長(zhǎng)因子(Endothelial Cell Growth Factor, ECGF)或肝素結(jié)合生長(zhǎng)因子(Heparin-Binding Growth Factor, HBGF-1),是一種分子量在17-18kDa之間的非糖 基化多肽,由多種細(xì)胞分泌產(chǎn)生。FGF-acidic分子具有155個(gè)氨基酸,不存在明顯的信號(hào)肽序列,因此排除了FGF-acidic以 經(jīng)典的ER/Golgi通路分泌的可能。FGF-acidic可能具有一種特殊的分泌方式,并且需要熱休克蛋白、磷酯酰絲氨酸、突觸結(jié)合蛋白1和膜聯(lián)蛋白的參與。FGF-acidic以二聚體的形式分泌,還原劑能夠破壞分子間的共價(jià)鍵,從而釋放具有生物活性的FGF-acidic。與FGF-2不同,F(xiàn)GF-acidic不具有可變的5’端轉(zhuǎn)錄起始位點(diǎn),目前發(fā)現(xiàn)的FGF-acidic剪切變體是一種氨基酸長(zhǎng)度為66的變體,其能與FGF受體結(jié)合但不能激活受體,具體的生物功能尚不明確。全長(zhǎng)形式的FGF-acidic具有一個(gè)氨基酸長(zhǎng)度在21-27之間的細(xì)胞核定位序列。表達(dá)FGF-acidic的細(xì)胞主要有乳腺上皮細(xì)胞、神經(jīng)細(xì)胞、骨骼及平滑肌細(xì)胞、腎近端小管上皮細(xì)胞、內(nèi)皮細(xì)胞、巨噬細(xì)胞、角質(zhì)細(xì)胞和成纖維細(xì)胞。
FGF家族共有5個(gè)相應(yīng)受體(FGF R1-R5),其中FGF R1-R4都屬于免疫球蛋白家族(Ig-Superfamily, IgSF) I型跨膜酪氨酸激酶受體,而FGF R5雖然也屬于免疫球蛋白家族成員,但缺少酪氨酸激酶區(qū)域。FGF-acidic主要與FGF R1-R4結(jié)合。FGF-acidic與受體結(jié)合后能夠激活酪氨酸激酶介導(dǎo)的信號(hào)通路,F(xiàn)GF和肝素之間的相互作用既能激活受體也能抑制受體,當(dāng)表現(xiàn)為激活作用時(shí),肝素主要是作為FGF與FGF R之間的中心連接物而存在。FGF-acidic也能與細(xì)胞表面的肝素單獨(dú)結(jié)合,此時(shí)只能誘導(dǎo)發(fā)生內(nèi)化作用而并不存在細(xì)胞活化作用。

FGF-acidic能夠促進(jìn)內(nèi)皮細(xì)胞發(fā)生有絲分裂,無(wú)論是細(xì)胞膜表面受體激活還是內(nèi)化作用對(duì)完整的有絲分裂應(yīng)答都必不可少。另外,F(xiàn)GF-acidic還能促進(jìn)平滑肌細(xì)胞、肝細(xì)胞、乳腺上皮細(xì)胞和成纖維細(xì)胞增殖。在發(fā)育和再生過(guò)程中FGF-acidic同樣具有重要作用。FGF-acidic能夠通過(guò)Notch-Delta通路抑制神經(jīng)元細(xì)胞的分化,誘導(dǎo)垂體前葉泌乳素細(xì)胞分化,促進(jìn)受損外周神經(jīng)軸突再生。另外,在CD4+ T細(xì)胞增殖過(guò)程中,F(xiàn)GF-acidic還是重要的共刺激分子,能夠促進(jìn)IL-2的分泌。
本試劑盒采用雙抗體夾心ELISA法(Sandwich ELISA)檢測(cè)樣品中小鼠FGF-acidic的濃度,其原理見(jiàn)圖1。小鼠FGF-acidic特異的單克隆捕獲抗體已預(yù)包被于酶標(biāo)板上,當(dāng)加入標(biāo)準(zhǔn)品或樣品時(shí),其中的小鼠FGF-acidic會(huì)與捕獲抗體結(jié)合。當(dāng)加入生物素化的抗小鼠FGF-acidic抗體后,生物素化抗小鼠FGF-acidic抗體與小鼠FGF-acidic結(jié)合,形成夾心的免疫復(fù)合物。隨后加入辣根過(guò)氧化物酶標(biāo)記Streptavidin (HRP-Streptavidin),由于生物素與鏈霉親和素(Streptavidin)可以特異性地結(jié)合,因此鏈霉親和素連接的HRP就會(huì)與夾心的免疫復(fù)合物連接起來(lái)而被固相捕獲。最后加入顯色劑TMB溶液,固相捕獲的辣根過(guò)氧化物酶就會(huì)催化無(wú)色的顯色劑氧化成藍(lán)色物質(zhì),在加入終止液后呈黃色。通過(guò)酶標(biāo)儀檢測(cè)450nm處的吸光度值就能實(shí)現(xiàn)定量檢測(cè)。小鼠FGF-acidic濃度與A450值呈正比,通過(guò)繪制標(biāo)準(zhǔn)曲線,對(duì)照樣品吸光度值,即可計(jì)算出樣品中靶蛋白濃度。

成纖維細(xì)胞生長(zhǎng)因子21 (Fibroblast growth factor 21, FGF21)屬于FGF家族,根據(jù)結(jié)構(gòu)特性其屬于FGF-19亞族,該亞族中還包括FGF-19和FGF-23成員。FGF家族成員大都具有一個(gè)氨基酸長(zhǎng)度為120的核心FGF區(qū)域,該區(qū)域具有β-trefoil結(jié)構(gòu)。FGF19亞族成員與其他FGFs不同,其并不含有β-trefoil結(jié)構(gòu),并且也不能與肝素等細(xì)胞外基質(zhì)分子牢固結(jié)合。因此,F(xiàn)GF-19亞族成員與其它FGFs相比更容易擴(kuò)散。FGF-19亞族成員利用輔助受體klotho家族直接與FGF受體結(jié)合,參與調(diào)控代謝相關(guān)過(guò)程。FGF-21利用C端序列與β-Klotho結(jié)合,從而傳遞信號(hào)。成熟小鼠FGF-21與成熟的人和大鼠FGF-21的同源性分別為81%和92%[1]。
當(dāng)PPARα/RXR復(fù)合物受到游離脂肪酸刺激后,會(huì)刺激肝細(xì)胞產(chǎn)生FGF-21。FGF-21分泌后,會(huì)誘導(dǎo)白色脂肪組織分泌游離脂肪酸,隨后游離脂肪酸在肝臟發(fā)生氧化,最終產(chǎn)生乙酰輔酶A,隨后乙酰輔酶A與一些4碳酮結(jié)合,參與能量生成。另外在脂肪組織中,F(xiàn)GF-21與PPARγ協(xié)同作用,產(chǎn)生更多的葡萄糖轉(zhuǎn)運(yùn)體,從而增加葡萄糖攝取。FGF-21能夠穿過(guò)血腦屏障,并造成精神遲緩,缺乏活力,在禁食或冬天時(shí)這會(huì)造成類似于冬眠的現(xiàn)象出現(xiàn)。在db/db和ob/ob小鼠中,F(xiàn)GF-21能夠調(diào)整葡萄糖和甘油酸三酯的值恢復(fù)到接近正常的水平,并增加胰島素的敏感性[2,3]。

Background:

FGF-10 belongs to the subgroup of FGFs that also includes FGF-3, -7, and -22. FGF-10 is secreted by mesenchymal cells and associates with extracellular FGF-BP. It preferentially binds and activates epithelial cell FGF R2 (IIIb) and interacts more weakly with FGF R1 (IIIb) [1]. The expression and function of FGF-10 are negatively regulated by Shh and BMP-4 in the developing lung [2, 3]. Overlapping expression patterns and activities with FGF-3, -7, and -8 suggest at least a partial redundancy in FGF?10 biology. FGF-10 induced signaling through FGF R2 (IIIb) also contributes to the progression of pancreatic cancer [4] .

Background:

Basic fibroblast growth factor (bFGF), also known as FGF2, is a member of the FGF superfamily of mitogenic proteins which show 35-60% amino acid conservation[1]. FGF acidic and basic are unique from other members of the family in that they lack classical secretory signal peptides. It is a highly specific chemotactic and mitogenic factor for many cell types, appears to be involved in remodeling damaged tissue, such as ulcer healing, vascular repair, traumatic brain injury[2]. FGF basic may play a role in vivo in the modulation of such normal processes as angiogenesis, wound healing and tissue repair, embryonic development and differentiation, and neuronal function and neural degeneration. Additionally, FGF basic may also participate in the development of several pathological conditions resulting from excessive cell proliferation and/or angiogenesis[3]. BFGF is a critical component of human embryonic stem cell culture medium[4].

DESCRIPTION

Background:

FGF-7 (fibroblast growth factor-7) is one of 22 known members of the mouse FGF family of secreted proteins that plays a key role in development, morphogenesis, angiogenesis, wound healing, and tumorigenesis[1].FGF-7 promotes cell migration and invasion, and mediates melanocyte transfer to keratinocytes upon UVB radiation[2]. It has been used ectopically to avoid chemotherapy-induced oral mucositis in patients with hematological malignancies. Deletion of FGF-7 affects kidney development, producing abnormally small ureteric buds and fewer nephrons[3] . It also impedes hair follicle differentiation [4]. The 194 amino acid (aa) FGF-7 precursor contains a 31 aa signal sequence and, like all other FGFs, an ~120 aa beta -trefoil scaffold that includes receptor- and heparin-binding sites.
